Kevin Glenn got the start under center for Calgary (5-1) and completed 21- of-28 passes for 218 yards and a touchdown in the Stampeders' fourth straight victory.
Nik Lewis led all receivers with seven catches for 96 yards and a score, helping the Stamps move into a tie with the Roughriders atop the Western Conference.
Darian Durant, who missed Saskatchewan's 32-20 triumph over Hamilton on July 27, returned under center and finished 20-of-32 for 278 yards and three scores, but was sacked three times in defeat.
Kory Sheets rushed for 133 yards on 14 carries and has racked up at least 130 yards in every game this season for the Roughriders (5-1), who defeated Calgary 36-21 in Week 2.
Sheets fumbled the ball on Saskatchewan's first touch and the Stamps needed just one play to convert the turnover into points, as Cornish took a direct snap and scampered 14 yards around the right end to make it 7-0.
After a quick two-and-out from the Roughriders, Cornish capped a lengthy scoring drive with a 2-yard TD surge. The extra point was blocked, leaving Calgary with a 13-0 cushion.
Rene Paredes poured in a 27-yard field goal early in the second to extend the lead before Durant got the Riders on the board with a 20-yard touchdown pass to Chris Getzlaf to make it 16-7.
Cornish broke free for a 32-yard major later in the second before Paredes added a 30-yarder on the last play of the half to make it 26-7 at the break.
Chris Milo knocked down a 28-yard field goal on Saskatchewan's first touch of the second half and after a two-and-out from Calgary, Durant found Getzlaf in the left corner of the end zone for a 3-yard major to make it 26-17.
Milo buried a 13-yard field goal with under a minute left in the third quarter to trim Saskatchewan's deficit to 26-20, but Durant was sacked in the end zone for a safety early in the fourth to push the Stamps' cushion to eight.
On the ensuing drive, Glenn hooked up with Lewis on three straight plays, including a 25-yard touchdown connection to make it 34-20.
Durant answered with a 23-yard scoring strike to Jock Sanders, but Cornish capped Calgary's next possession with a 1-yard TD plunge to put the game away.
Paredes connected on both of his field goal tries to push his record streak of consecutive made field goals to 36 ... Milo went 2-for-2 on field goal tries and is a perfect 20-for-20 on the season ... Getzlaf finished with three catches for 43 yards and a pair of touchdowns.
